In this lab, students use classic biochemical techniques to isolate the protein sucrase from yeast cells. The identity of the isolated protein, an enzyme, is verified by testing its activity in the specific digestion reaction catalyzed by sucrase. Students learn about the properties of proteins and enzymes, how biologists identify different proteins, and the unique roles of proteins in the life of a cell. Enough material for 30 students working in pairs. Requires a centrifuge.
